Glucosylated resveratrols were isolated from crude methanol extract of Polygonum cuspidatum by semi-preparative high speed countercurrent chromatography (HSCCC, for details see cited literature). The HSCCC fractions were suspended in 1 mL 5 % methanol and purified by SPE.

Conditions

Column conditioning: 6 mL methanol, then 6 mL water (Do not let run the column dry.)

Sample aspiration: The pretreated sample is loaded onto the column

Washing: 6 mL 5% methanol

Elution 1: 18 mL 30 % methanol Elution 2: 12 mL 50 % methanol Elution 3: 6 mL methanol

Evaporation: to dryness of the seperately collected fractions

HPLC-DAD and HPLC-ESI-MS with C18 column (e.g., EC 150/3 NUCLEODUR C18 Gravity, 5 µm, REF 760103.30), conditions see cited literature 1H-NMR, HMBC-NMR
