Eluent : acetonitrile - water (50:50, v/v)
Flow rate: 0.56 mL/min
Temperature: 25 °C
Injection volume: 1.0 µL
Detection
UV, 254 nm
Note
The separation of pyridine and phenol is used to determine the base deactivation. On a good base deactivated phase pyridine is eluted well ahead of phenol. Furthermore the pyridine peak has to be symmetric. NUCLEODUR PolarTec shows a high resolution between pyridine and phenol, and the pyridine peak is symmetric. This indicates a very good base deactivation.
